 OBJECTIVE: To establish the determination method for complanatoside A in seeds of Astragalus complanatus. METHOD: An HPLC method has been developed to separate complanatoside A on ZORBAX EXTEND-C18 (4.6 mm x 250 mm, 5 microm) column with acetonitrile-water-phosphoric acid (20:80:0.2) as mobile phase and UV detection at 267 nm. RESULT: The good linearity of complanatoside A ranged 0.086-0.430 microg, r = 0.9999. An average recovery of 99.8% (n = 5) was obtained with a RSD of 1.0%. CONCLUSION: The established method is proved to be stability, fast, accurate and can be used for quantification of Complanatoside A in Semen Astragali Complanati.

 AIM: To observe the anti-liver fibrosis effect of Astragalus complanatus flavonoids (ACF) in rats. METHODS: The liver fibrosis model in rats was established by injecting interperitoneally 0.2 mL/100 g 0.5% dimethylnitrosamine, thrice a week. Meanwhile, the rats were administered ACF (30, 60, 120 mg/kg) or colchicine (0.1 mg/kg) once a day for 1 mo. Serum N-propeptide of type I procollagen (PINP) and type III procollagen (PIIINP) were measured using ELISA. Malondialdehyde (MDA) and superoxide dismutase (SOD) in hepatic tissue were evaluated. Matrix metal protease-1 (MMP-1) mRNA expression was assayed by RT-PCR and the protein expression of tissue inhibitor of metal protease-1 (TIMP-1) was analyzed by immunohistochemistry. RESULTS: In the ACF groups, SOD activity increased and MDA content decreased in comparison to the liver fibrosis model group. The serum PINP and PIIINP contents in ACF-2 and -3 group decreased compared to those in model group. In ACF-2 and -3 group, the expression of MMP-1 mRNA increased significantly and the protein expression of TIMP-1 decreased compared to that in model group. CONCLUSION: The antifibrotic mechanisms of ACF are associated with its influence on lipid peroxidation and collagen synthesis and degradation.

 The purpose of the present study was to quantify the antihypertensive effect of the total flavonoid (TF), extracted from the seed of Astragalus complanatus R. Brown, and to observe its effect on the renin-angiotensin system (RAS) in both renal hypertensive rats (RHR) and spontaneously hypertensive rats (SHR). RHR were created by the two-kidney one clip (2K1C) method. Systolic blood pressure was measured in conscious rats by the tail-cuff method. Plasma angiotensin II (AngII) and plasma renin activity (PRA) were measured with radioimmunoassay at 60 min after drug administration. The effects of TF on cardiac hemodynamics were also recorded in anesthetized RHR and SHR. TF was given by oral administration in low dose (100 mg/kg) and high dose (200 mg/kg) respectively. Compared to pre-administration control, TF induced an obvious decrease in systolic blood pressure in conscious normotensive Wistar rat, RHR and SHR. In the three groups the systolic blood pressure reached the lowest value at 60 min after TF. TF also induced a significant decrease in blood pressure in anesthetized RHR and SHR. At 60 min after treatment of TF, mean arterial pressure in high dose group (200 mg/kg) was decreased by 17% in RHR and by 17% in SHR respectively (P < 0.01). The depressor effect of TF lasted for at least 60 min. Cardiac output, heart rate and +/- dp/dtmax did not change. Conversely, total peripheral resistance was significantly decreased. The decrease in plasma AngII was found in both RHR and SHR. On the contrary, PRA increased at the same time. These findings suggested that TF is effective in reducing blood pressure in both RHR and SHR. The antihypertensive action of TF was attributed to a decrease in TPR secondary to a decrease in plasma concentration of AngII caused by TF.

 OBJECTIVE: To establish the identification method for genuine and pseudo Shayuanzi by HPLC fingerprint. METHOD: The flavone, a kind of active composition in genuine (Astragalus complanatus) and analysed species was analysed by RP-HPLC. RESULT: The fingerprints of genuine Shayuanzi collected from different places showed the similar characteristics, but they were different from those of pseudo species A. adsuragen and A. sinicus. CONCLUSION: The HPLC fingerprint can be an identifing method for Shayuanzi.

 OBJECTIVE: To understand the varieties of Shayuanzi on sale and establish the method for evaluating its quality. METHOD: 22 samples collected from all over the country were identified by TLC and the contents of total flavone in them were determined with UV. RESULT: All samples could be classified into three varieties, and they could be identified by TLC. The contents of total flavone in them were 0.3%-1.0%. CONCLUSION: Shayuanzi on sale mainly includes Astragalus complanatus, and there are A. adsuragen and A. sinicus as well. TLC is a better method for the identification of shayuanzi. The total flavone in A. complanatus is 0.33%-0.82%.

 OBJECTIVE: To investigate effects of total flavonoid fraction of Asttragalus complanatus on blood pressure in conscious spontaneously hypertensive rats(SHR) and hemodynamics in anesthetized SHR. METHOD: Blood pressure was recorded with tail-culf method and hemodynamics was recorded with polygraph after administration of total flavonoid fraction of Asttragalus complanatus. RESULT: Total flavonoid fraction of Asttragalus complanatus (100, 200 mg.kg-1) could decrease blood pressure of conscious SHR(decreasing 7.1%, P < 0.05 and 9.3%, P < 0.01 respectively). Meanwhile, there was no significant change in heart rate(HR). R Br (200 mg.kg-1) could decrease total peripheral resistance (TPR) (decreasing 20%, P < 0.05) but did not influence the cardiac output(CO) and heart rate in anesthetized SHR. CONCLUSION: Total flavonoid fraction of Asttragalus complanatus possesses obvious hypotensive effect, mainly by means of decreasing the TPR.

 Micrograph were used to analyse the number, karyotype and volume of chromosome of Astragalus complanatus. The normal diploid 2n = 16, karyotype formula based on Levean publication (1964) was K(2n) = 16 = 10 m + 6 sm. According to the method of S.R. Guo, the chromosome relative length was 2n = 16 = 6M2 + 10M1, which belonged to "2A" type according to the Stebbins' karyotype classification. The total length of chromosome groups was 33.05 microns, total length of long arms was 19.76 microns, from Arano's method the AS.K% was 59.79%. The total volume of chromosome was 50.6 micron 3.

  Study of structure--hepatoprotective relationships of oleanene-type triterpenoidal glucuronides obtained from several fabaceous plants on rat primary hepatocyte cultures.:Biol Pharm Bull. 1999 Feb;22(2):203-6.

 The protective effects of nine oleanene-type triterpenoidal glucuronides obtained from several fabaceous plants (Lupinus polyphyllus x arboreus Hybrid, Astragalus complanatus, Wistaria brachybotrys) on in vitro immunological liver injury in primary cultured rat hepatocytes were studied. Although all tested saponins showed hepatoprotective action, the levels of activity of the individual saponins differed. Structure-activity relationships for the sapogenol moiety suggested that the presence of the carbonyl group at C-22 would show a similar hepatoprotective effect to that of the hydroxyl group at C-22 while the presence of the hydroxyl group at C-30 would reduce the hepatoprotective action. This structure-activity relationship substantiated other recently obtained data. Furthermore, the beta-orientation of the hydroxyl group at C-21 is more effective than the alpha-configuration in regard to the hepatoprotection. Furthermore, structure-activity relationships for the sugar moiety substantiated previously obtained data that the hydroxyl group at C-5" enhances the hepatoprotective action.

 The petroleum ether extract of Astragalus complanatus R. Br. showed activity for lowering the transaminase levels in mice. From this extract six compounds were isolated. Among them five were identified as rhamnocitrin (I), formononetin (II), stigmasterol (IV), phosphatidylethanolamine (V) and phosphatidylinositol (VI). The other one was shown to be a new p-coumaroyl derivative, 11-O-p-coumarylnepeticin. Their structures were elucidated by spectrometric and chemical analyses. All the six compounds were isolated from this plant for the first time.

 beta-sitosterol and fatty acids were obtained from the oil of seeds of Astragalus complanatus. The fatty acids were separated and their molecular weights determined by GC-MS. Fourteen of them were identified as heptenoic acid, tetradecanoic acid, pentadecanoic acid, hexadecanoic acid, octadecenoic acid, octadecanoic acid, octadecadienoic acid, linolenic acid, eicosanoic acid, eicosenoic acid and docosanoic acid, etc.

 Sha-Yuan-Zi is the seeds of Astragalus complanatus R. Br.. Eight compounds were isolated from its 80% ethanolic extract. Their structures have been identified by spectroscopic and chemical methods. Two of them are new and named neocomplanoside (VII) and myricomplanoside-(VIII) respectively. All the compounds were obtained from Sha-Yuan-Zi for the first time.
