Lobeline has a long history of therapeutic use ranging from an emetic and respiratory stimulant to a tobacco smoking cessation agent. Lobelia chinensis Lour, a traditional Chinese herb whose active ingredient is the alkaloid lobeline, demonstrated to antagonize the bioactive effect of endothelin-1 (ET-1) and prevent the proliferation of vascular smooth muscle cells (VSMCs) in hyperlipidemic rats. The objective of the present study was to determine the effects of lobeline on proliferation of cultured human umbilical VSMCs induced by ET-1. The results showed that the increased cell numbers and enhanced [3H]thymidine incorporation induced by ET-1 were inhibited and the transition of cells from static phase (G0/G1) to DNA synthesis (S) and mitotic phase (G2/M) was held back by lobeline in a concentration-dependent manner. Confocal microscopy demonstrated that lobeline markedly decreased the fluorescent intensity of intracellular Ca2+ concentration ([Ca2+]i) with a significant difference from ET group. Cytotoxicity was determined by Trypan blue exclusion. These results demonstrated a novel biological role of lobeline. Lobeline inhibited the proliferation of human umbilical VSMCs induced by ET-1 in a dose-dependent manner and the anti-proliferative effect was involved in the reduce of increased [Ca2+]i, rather than nonspecific cytotoxicity.

OBJECTIVE: To develop an HPLC method for determination of two polyacetylenes, lobetyolin and lobetyolinin, in Herba Lobeliae Chinensis. METHOD: C18 column was used with the mobile phase consisted of acetonitrile and water. Linear gradient elution from 10% to 40% acetonitrile in 25 min was applied, at the flow rate of 1.0 mL x min(-1), the detection wavelength was at 267 nm. RESULT: Lower contents of lobetyolin and lobtyolinin were found in collected samples of Herba Lobeliae Chinensis. The highest amounts of lobetyolin and lobetyolinin were found to be 0.461 and 0.436 mg x g(-1) in a sample procured from Hong Kong. However, there were no lobetyolin and lobetyolinin in some of the samples. CONCLUSION: A simple and effective HPLC method to analyze the two polyacetylenes in Herba Lobeliae Chinensis was established. It could be applied for the quality control of this herb.

The effects of a series of substances on the biological function of endothelin (ET) are reported. The substances used are: synthetic inhibitors of endothelium derived relaxing factors (EDRFs), inhibitor of big-endothelin converting enzyme phosphoramidon, antiserum of endothelin, antagonists of endothelin A receptor BQ123 and JKC301, and two Chinese anti-snake venom herb medicines Lobelia radicans Thumb and Taris polyphylla Smith var. chinensis (Franch) Hara. The results showed that inhibiting the production of nitric oxide (NO) could stimulate ET release from vascular endothelium, elevate plasma ET and increase blood pressure. These changes could be reversed by L-arginine (L-Arg), the substrate of nitric oxide synthase (NOS). The amount of ET released by arterial endothelium could be increased or inhibited by inhibiting or stimulating the synthesis of prostacyclin (PGI2). The plasma ET level and blood pressure in both SHR and WKY rats could be decreased by giving phosphoramidon (PhR). The above results indicate that the biological effects of ET could be antagonized by inhibiting the synthesis or release of ET, decreasing the level of plasma ET, blocking the binding of ET with its receptor and using some Chinese anti-snake venom herb medicines.
