Emodin is a naturally occurring anthraquinone present in the roots and bark of numerous plants of the genus Rhamnus. Extracts from the roots, bark, and/or dried leaves of buckthorn, senna, cascara, aloe, frangula,and rhubarb have been used as laxatives since ancient times and currently are widely used in the preparation of herbal laxative preparations. Anthraquinone glycosides are poorly absorbed from the gastrointestinal tract but are cleaved by gut bacteria to produce aglycones (such as emodin) that are more readily absorbed and are responsible for the purgative properties of these preparations. There is extensive exposure to emodin and other anthraquinones resulting from the use of herb-based stimulant laxatives. Reports that 1,8-dihydroxyanthraquinone, a commonly used laxative ingredient, caused tumors in the gastrointestinal tract of rats raised the possibility of an association between colorectal cancer and the use of laxatives containing anthraquinones.

Because emodin is a hydroxyanthraquinone structurally similar to 1,8-dihydroxyanthraquinone,is present in herbal laxatives,and was reported to be mutagenic in bacteria, it was considered a potential carcinogen and was selected for in-depth evaluation. Male and female F344/N rats and B6C3F1 mice were exposed to emodin (at least94% pure) in feed for 16 days, 14 weeks, or 2 years.
Genetic toxicology studies were conducted in Salmonella typhimurium, cultured Chinese hamsterovary cells, rat and mouse bone marrow cells, and mouse peripheral blood erythrocytes.
Emodin (3-methyl-1,6,8-trihydroxyanthraquinone) inhibits TNF-induced NF-B activation, IB degradation, and expression of cell surface adhesion proteins in human vascular endothelial cells
Most inflammatory agents activate nuclear transcription factor-B (NF-B) which results in expression of genes for cytokines, adhesion molecules, and enzymes involved in amplification and perpetuation of inflammation. Emodin (3-methyl-1,6,8-trihydroxyanthraquinone) is an active component from the roots of Polygonum cuspidatum that has been reported to exhibit antiinflammatory properties but the mechanism is not known. In the present study we investigated the effects of emodin on the activation of NF-B in human umbelical vein endothelial cells (EC). Treatment of EC with TNF activated NF-B; preincubation with emodin inhibited this activation in a dose- and time-dependent manner. Emodin did not chemically modify NF-B subunits but rather inhibited degradation of IB, an inhibitory subunit of NF-B. Since the promoter regions of ICAM-1, VCAM-1, and ELAM-1 contain NF-B binding sites and these adhesion molecules are involved in the attachment of leukocytes to EC, the effect of emodin on the adhesion of monocytes to EC and the expression of these adhesion molecules was also studied. Treatment of EC with TNF for 6 h increased the adhesion of monocytes to EC, which correlated with increases in cell surface expression of ICAM-1, VCAM-1 and ELAM-1. Pretreatment of EC for 1 h with emodin inhibited both monocyte-EC attachment and expression of ICAM-1, ELAM-1 and VCAM-1.
These results indicate that emodin is a potent inhibitor of NF-B activation and expression of adhesion molecules and thus could be useful in treating various inflammatory diseases.

A recent study by Tufts University and Korean investigators revealed a dramatic 88% drop in breast cancer in women who had the highest blood concentrations of lutein.
Lutein and its oxidative metabolites in chemoprevention of cancer:
Carotenoids are abundant in fruits and vegetables and have been proposed as cancer protective compounds because of their antioxidant and provitamin A activities.
Lutein, an abundant carotenoid in many fruits and vegetables but without vitamin A activity, has been shown to possess strong antioxidant capability in laboratory studies. Results from the present clinical study from Beltsville demonstrate that lutein and zeaxanthin, an isomer of lutein, are partially oxidized in vivo to several metabolites. These observations confirm the antioxidant activity of lutein in vivo which supports one of the proposed cancer protective mechanisms of carotenoids. Results and conclusions from these studies will benefit the diet and health community as well as policy makers.
Lutein as cancer inhibitor:
Researchers at the University of Utah Medical School found that the highest consumers of lutein (a mere 2.4mg daily) were 17% less likely to develop colon cancer than those who ate the least (300 micrograms). Generally, the more lutein consumed, the lower the risk. High lutein also has been linked to fewer lung, prostate and ovarian cancers.
In animals, lutein even slowed the growth of breast tumors. In test tubes, it killed cancer cells.

Mice Experiment of Lutein Against mammary tumor
Mice were fed a diet containing 0.1% or 0.4% of lutein.
In experiment 1, mice were fed on the diets for 3 weeks and infused with mammary tumor cells into the mammary gland. Result show that dietary lutein increased tumor latency and inhibited mammary tumor growth in a dose-dependent manner. The incidence of palpable tumors on day 28 post-infusion and final tumor weight were lower in mice fed lutein.
In experiment 2, dietary lutein enhanced phytohemagglutinin-induced lymphocyte proliferation but had no effect on interleukin-2 production or lymphocyte cytotoxicity. Therefore dietary lutein increased tumor latency, suppressed mammary tumor growth and enhanced lymphocyte proliferation.

Antioxidant and Anti-carcinogenic Effects:R. rosea is rich in phenolic compounds, known to have strong antioxidant properties.53,86 Animal studies have shown that R. rosea decreases toxicity from cyclophosphamide, rubomycin, and adriamycin (anti-cancer drugs), while it enhances their anticarcinogenic effects.87-89 Udintsev and Schakhov studied the effect of R. rosea root extract (RRRE), a tincture manufactured according to the Russian Pharmacopoeia standards (minimum 0.8 percent salidroside and 3 percent rosavin), on tumor cells (transplanted into mice) and normal bone marrow cells in two mouse cancer models.90 One group of mice with Ehrlich ascites tumor (EAT) and another group with Lewis lung carcinoma (3LL) were first treated with 100 mg/kg cyclophosphamide (a chemotherapy agent) that suppressed tumor growth to 31-39 percent and limited 3LL metastases to 18 percent, while also reducing the number of normal bone marrow cells, leucocytes, and myelokariocytes, to 40-50 percent and 20-25 percent of normal, respectively. In comparison, RRRE, 0.5 mg/kg/day given orally 2-8 days after tumors had been transplanted, suppressed growth of both tumors by 19-27 percent and 3LL metastases 16 percent. However, in contrast to cyclophosphamide, RRRE caused no reduction in normal bone marrow cells. In animals given both RRRE and cyclophosphamide, the RRRE increased the antimetastatic effect of cyclophosphamide by 36 percent (p<0.05). RRRE also increased the number of leukocytes by 30 percent and myelokariocytes by 16-18 percent.
